Everyone has an opinion, so let's do a simple poll, for those who bike a lot.
forum name/gender/saddle make & type/ love it, hate it, or suffer it

Blisters/male/ Specialized BG Riva/ suffering it

All saddles are horrid. 

My rambling experience

Kuota standard. Nah, not even there.
Profile Design tri - chafes inside leg, soft but not a long distance saddle
Specialised BG tritip gel - Fucking brick
Selle San Marco Triathgel Aspide - OKish but VERY bad sacro pain on left side
Selle Italia new leather anti numb knob one - Jury out but leather is good, still lots of lower back pain, costing me a fortune in Dubbin, knob still numb.

Conclusion - natural materials ie leather will be the way to go.

Dubai Dave / Male / Fizik tri / Its OK

Road Bike / Selle Italia SLR / Its Ok

Tried Specialized Toupe / Awful

ISM Idamo / ended up with blisters on thighs

Selle Italia  Tri / Its OK use on TT seat post on my road bike

In my opinion it makes no odds, first hour is OK, 2nd hour arse hurts, after 2 hours the level of pain gets no worse, numbness sets in / brain turns off

Road bike Selle I talia SLR minimalist but very comfy over 100 miles
Tri bike planet x titanium ( the fizik copy) great over IM distance in the aero position
Cross/tourer Brooks Team Pro the comfiest saddle ever if it didnt weigh half a ton and look like it was designed in 1066 i would have it on all my bikes
MTBs charge spoon nice n comfy bit of padding and cleans up well

road bike...........the planet x one that came with my bike so a male model.......i can sit on it for hours.infact 13 hours in one night  last month and no problems at all.......

I am female aand love it

TT Bike / Specialized Romin Comp / loved it form day 1!

Road bike / Specialized Avatar / Its OK - has more padding than ther Romin, but is less comfortable!

planet-x superlight is the best i've tried. I've never had any major discomfort on it even in a tri suit for a few too many hours. I have a selle italia shiver on my road bike and even with well padded shorts its not great.

Fizik arione- on the whole its fab, but I must remember to get out of the saddle now and again (serious knob numbage post IMCH as I did nearly all the climbs seated).

Boardman saddle, despite a similar profile to the fizik- hated it.

Another vote for the Planet X superlight although I've just got a Charge Spoon so am trying that out for a bit & seems OK so far.

Fizik Arione on race and training bike - comfort like no other saddle

Charge Spoon on singlespeed - for value for money and comfort this is probably the best saddle available today.  - less than £20 on Wiggle and of the near 500 reviews, most are very positive.  most negatives relate to colour choice which is not a comfort issue!!  it's quite a bargain and it's worth spending that amount just to try it
